Vector Laboratories - Reducing Tissue Autofluorescence: Innovations in Immunofluorescence Analysis Boost Signal-to-Noise Ratios

Immunofluorescence can provide clear subcellular visualization of proteins, glycans, and small biological and nonbiological molecules. However, tissue autofluorescence often impairs or prevents the effective use of this tool. As will be described, a new technology has been developed that significantly lowers autofluorescence and dramatically enhances signal-to-noise ratios.

Micronic - How to Standardize Tissue Sample Storage

The validity of the data of a research project depends on the quality of the tissue samples. Therefore, it’s important that a tissue storage facility preserves the quality of its samples. Variations in tissue sample storage conditions will lead to variations in the quality and characteristics of the tissue samples. To prevent quality alterations, strict standards in tissue banking are needed. Understanding the potential of the research on tissue samples leads to the conclusion that the standardization of biobanking practices are of utmost significance.

Associates of Cape Cod – Variability and Test Error with the LAL Assay

American Pharmaceutical Review™ article re-publication.

The Limulus Amebocyte Lysate (LAL) assay is the compendial test for the examination of bacterial endotoxin in pharmaceutical products (as described in USP chapter <85>), in-process material, and pharmaceutical grade water.

With any biological tests, measurements are susceptible to variations in analytical conditions.  Here the LAL assay has a relatively high level of variability even for a biological assay.  This variation derives from 3 principle sources: reagents, product, and method.  This paper examines some of the reasons for LAL test variation, focusing on photometric methods (chromogenic and turbidimetric), and considers how variation can be assessed through good laboratory quality control.

Mirus - Optimization of DNA, RNA and RNP Delivery Methods for Efficient CRISPR/Cas9 Mediated Cell Engineering

The success of CRISPR genome editing experiments is limited by the intracellular delivery and expression of Cas9 Protein and gRNA. Many methods for achieving CRISPR mediated cleavage have been identified and the choice of DNA, RNA or ribonucleoprotein (RNP) format is dictated by experimental goal and cell type. Transfection of each type of molecule requires specific considerations for efficient functional delivery. We performed transfections using different combinations of molecules including: plasmid DNA, messenger RNA, Cas9 protein and gRNA to maximize targeting of the Cyclophilin B (PPIB) gene in HEK293 T/17 and other mammalian cell types. Their results extend the utility of the CRISPR/Cas9 system by identifying optimal transfection conditions for intracellular delivery of Cas9 and gRNA in different formats.

EpiCypher - Nucleosome Remodeling Assay by EpiDyne™-FRET

Chromatin  remodeling, or the repositioning of nucleosomes, regulates DNA access and thus gene expression and genome repair. Many ATP-dependent remodeling enzyme complexes are associated with human disease but are challenging study targets due to the requirement for nucleosome-based substrates.  EpiCypher has addressed this need by developing the EpiDyne platform of fully recombinant remodeling substrates to monitor nucleosome repositioning along DNA.  Here, we demonstrate the utility of EpiDyne-FRET in a homogeneous remodeling assay using Fluorescence Resonance Energy Transfer (FRET) readout.

Hycult Biotech - Complement in First Episode Psychosis

There has been debate over the past few years about the criteria currently used to classify patients with psychiatric disorders. The lack of objective and quantitative diagnostic criteria means that we rely on symptom-based categorization when deciding on patient classification and less on the underlying biology. Accumulating evidence suggests that inflammation and the immune system is the cause of such psychiatric disorders. The complement system is one such system, particularly the classical pathway, that potentially could be an important contributor.

Cygnus Technologies - New Methods Help Identify Individual Host Cell Proteins in Final Drug Product

Impurities can negatively affect the stability, safety, and efficacy of protein therapeutics.  Host cell proteins are a major part of process-related impurities during biologics production, and therefore must be carefully monitored and controlled. With Host Cell Protein ELISA being a critical component of ensuring process consistency and product purity, regulatory agencies have measures in place to ensure the Host Cell Protein ELISA used by a sponsor is fit for this purpose.  BioPharm International recently spoke with Ken Hoffman, president of Cygnus Technologies, about best practices and new technologies for host cell protein detection and identification.

Jena Bioscience - Crystallography: Impact of Crystal Mounting Method on Cryocooling & Diffraction

MiTeGen’s patented family of MicroMounts and MicroLoops are manufactured from a highly x-ray transparent polymer into precise shapes and sizes using cutting-edge photolithography techniques. These x-ray transparent loops can go directly into the beam, so users select a loop size that is slightly smaller than the crystal. They then easily harvest the crystal, which sits directly onto the surface of the loop. This reduces unwanted fluid around the crystal, leading to easier cryoprotecting, faster cryocooling, and less ice formation.
