ASSOCIATES OF CAPE COD INCORPORATED™ - Removal of Endotoxin from Protein in Pharmaceutical Processes

Bacterial endotoxin is the lipopolysaccharide component of the cell wall of gram-negative bacteria, together with other cellular material that combines to form an endotoxin complex.  Endotoxin is pyrogenic and it presents a risk to patients who are administered intravenous and intramuscular preparations.  Thus, bacterial endotoxins pose a risk to many pharmaceutical processes and, where not controlled, to the finished products.  There are different methods for endotoxin removal.  These include depyrogenation, such as dry-heat processes applied to glassware, and rinsing, as might be applied to closures.  These areas receive reasonable coverage within the pharmaceutical sector.  Areas that are actively discussed within biotechnology fields but which receive less attention in the wider pharmaceutical context are steps to remove endotoxin bound to protein.  This article considers some of the biotechnological applications for endotoxin removal.

abc biopply - Extracellular Vesicles Composition Affected by 3D Cell Architecture
 

abc biopply logo3D CoSeedis™ has recently been used to demonstrate the effect of 3D cellular architecture on microRNA and protein cargo of extracellular vesicles.  In a recent study, 3D CoSeedis™ was used to address the impact of cellular architecture on extracellular vesicle content and function. To do so, the authors used comparative analysis of biochemical features, RNA and proteomic profiles of vesicles secreted by both, 2D and 3D cultures of gastric cancer cells.

Hycult Biotech LogoLoonen, A et al; Soluble mannose receptor levels in blood correlate to disease severity in patients with community-acquired pneumonia. Immunology Letters 2018.

Community-acquired pneumonia (CAP) is the most common form of pneumonia and is a leading infectious cause worldwide. sMR, soluble mannose receptor, is a new serum marker for macrophage activation. Its extracellular domains are able to bind several ligands, like mannose, collagen and sulfated carbohydrates. Studies have shown that certain infection markers are (partly) capable to predict disease development in CAP. Since the need for new biomarkers to predict disease prognosis more precisely is still present, sMR was suggested. Results show that patients with sMR levels between 100 - 430 ng/ml had a 22.7% chance to develop severe disease, while patients with higher levels had a 33.3% chance.

EpiCypher – EpiDyne®: Exploring Chromatin Remodeling Substrates with Nucleosomes
 

EpiCypher Logo Nucleosomes are the fundamental repeating units of chromatin, comprised of a histone octamer wrapped by ~147bp DNA.  A common focus of epigenetics research is the addition / removal of various post-translational modifications (PTMs) to histone tails, and how these marks impact downstream processes, such as transcription, cell division, and disease.  Equally important, but less well understood, are the complexes that assemble, reformat, slide or eject nucleosomes in existing chromatin architecture.  These nucleosome remodeling complexes are large, multi-protein structures, which are powered by the hydrolytic activity of associated ATPases1.  Nucleosome remodeling is essential to the regulation of multiple pathways in cells, including transcription, genome replication, and DNA damage repair.
