Vector Laboratories – Immunohistochemistry Troubleshooting Tip

I am performing IHC on tissue sections using a mouse IgG primary antibody.  My detection system is a biotinylated anti-mouse IgG secondary antibody, a VECTASTAIN® Elite ABC peroxidase kit (VECTPK6100), in combination with a DAB substrate kit (VECTSK4100), and I do not see any staining. How do I know if the end detection reagents are working?

Make a fresh working solution of DAB substrate per instructions.  Place a small volume (~1 mL) of this DAB substrate into a clean glass test tube.  To this 1 mL, add one drop (~50 µl) of Reagent B only from the VECTASTAIN® Elite ABC kit.  If the HRP enzyme is active and the DAB reacts with it, an immediate color change will be observed.  This indicates the end detection reagents are working appropriately.

Associates of Cape Cod - Endotoxin Test Concerns of Biologics Part II: Developing New Tools

Bacterial Endotoxin Test (BET) users seek ways to overcome low endotoxin recovery (LER) from direct spikes into undiluted biologics.  These studies have come about from Chen’s initial observation that Control Standard Endotoxin (CSE) spikes, when placed into undiluted biologics, buffers, and other constituents often cannot be recovered.  By changing the endotoxin spike requirement from diluted product to undiluted product, users will unsurprisingly encounter test interference; what is unexpected from Chen’s LER finding is that recovery sometimes cannot be improved by dilution. This is indicative of a still undefined binding phenomenon.  Chen originally dismissed protein binding as a mechanism of LER as placebo with no protein of the same formulation also exhibited a low endotoxin spike recovery problem.  However, we should not assume that the binding phenomenon with protein and without protein demonstrate the same mechanism of spike loss.  This article will discuss a couple of different phenomena to explain LER observations; they may be dependent on both the protein concentration and the protein charge (anionic versus cationic) as well as surfactant and excipient effects.

NuGEN - Simple, Accurate, Molar Quantification of NGS Libraries

Accurate quantification of NGS libraries is required for proper loading of sequencing instruments and uniform read distribution across multiple samples.  Mass-based measurements such as UV absorbance or intercalating fluorescent dyes are not sufficient, since proper loading requires knowing the molar library concentration.  Users must estimate the average size of the library in order to calculate the molar library concentration.  This not only requires a separate fragment size analysis to be performed but also contributes to uncertainty especially in libraries with broader insert length distributions.  Here, NuGEN describes the performance of a novel library quantification method that produces a molar library concentration without the need for a separate size analysis.  The method can be performed with a simple fluorescent measurement, such as can be obtained with a standard fluorescent plate reader or Qubit instrument.  Excellent linearity was observed across a range of input concentrations, limited only by the dynamic range of the fluorescent reader.  NuGEN tested the method against a range of different average library sizes and size distributions and found good agreement with a popular real-time PCR based quantification method.  More importantly, they found that the method correlates well with cluster density on Illumina sequencers.

Antimicrobial peptides (AMPs) have historically been evaluated for their role in protecting against uropathogens. However, AMPs have not been evaluated for expression in noninfectious urological injury such as urinary tract obstruction. The objective of this pilot study is to compare urinary AMP expression in children undergoing surgical intervention for ureteropelvic junction obstruction (UPJO) with non-obstructed controls.
